Is this fuse going in between the alternator & battery, or battery & amplifier?

Are you using 1/0 wire from the battery to the amplifier?

Does the amplifier have on-board fusing?

250 will be fine! it is what I used back in the day for years becasue when I went to purchase 300A they were out and I never got around to buying a new one/ upgrading lol

If you use a 250 amp fuse, how will the alternators 300amps be able to flow through? The fuse will break.

Sounds like a bad time to me.
